Mr Musk has been chief executive of Twitter since paying $44bn for the company in a private capacity last year.
Since Musk bought Twitter, many longtime advertisers have abandoned the company, citing Musk's "axe swinging" and firing 80 percent of Twitter's staff, as well as concerns that their ads could appear next to inappropriate tweets due to a lack of censorship.
Earlier this year, Musk acknowledged that Twitter's online advertising revenue had fallen dramatically.
Musk said that Iaccarino could develop Twitter into an all-encompassing super client, offering diverse services such as personal money transfers. But media analysts say Musk's choice of a veteran of the advertising industry shows that online advertising will remain a key part of Twitter's future revenue model.
Musk has repeatedly said he is looking for a new chief executive to lead Twitter in his place. Mr Musk has limited energy as chief executive of electric car maker Tesla and private rocket launch company SpaceX.
After announcing the appointment of Twitter's new CEO, Musk said Acarino's arrival would allow him to devote more of his personal time to Tesla.
